Biosynthesis of collagenase by human skin fibroblasts in monolayer culture.
The biosynthesis of human skin collagenase was studied in fibroblast cultures incubated with 13H]leutine under serum-free conditions. The radioactively labeled enzyme was isolated and quantitated directly from culture medium using a highly specific immunoprecipitation method. Sodium dodecyl sulfate gel electrophoresis of the immunoprecipitates showed two radioactive bands tihich co-migrated wit...
متن کاملProduction of Collagenase Inhibitor by Mouse Calvaria in Tissue Culture
Acollagenase inhibitor was identified in the culture medium of mouse calvaria after separation from mouse bone collagenase by Sephadex G-200 gel filtration equilibrated with 6M urea in Tris buffer. The simultaneous synthesis of both collagenase and inhibitor by mouse calvaria in tissue culture was consistent with the resuls of our previous study using achick bone culture system and otehr recent...
متن کاملCollagenase in equine cell culture preparation.
Equine kidney cells disaggregated by treatment with 0.01% collagenase were used in the preparation of primary monolayer cell cultures. The primary cells could be stored for long periods in liquid nitrogen and subsequently subcultivated. These techniques provided a long-term supply of equine kidney cells, free of apparent contamination, from the kidneys of a single fetus.
متن کاملKeratoconus experimentally produced in mice using collagenase
Introduction: Keratoconus is a relatively common disease of cornea in which structural changes within the cornea cause it to thin and change to a conical shape and scar at the central portion of cornea. So far, few methods and drug treatments were introduced due to both lack of accepted animal models to induce experimental keratoconus and limitation of research in human considering ethical issu...
